Assessment of Acrylamide-induced Neurotoxicity Using Proteomics-based Technologies
A recent proteomic study provided evidence that N-ethylmaleimide sensitive factor (NSF), a rate-limiting ATPase involved in presynaptic transmitter release, was a target of in vivo or in vitro ACR exposure [1]. To confirm and extend these initial findings, liquid chromatography-tandem mass spectrometry (LC/MS/MS) was used to identify ACR protein adducts in tryptic digests of synaptosomes isolated from moderately ACR-exposed rats. Analyses using isotope-coded affinity tags (ICAT) revealed quantitative changes in the relative abundance of many cysteine-containing peptides (indicating potential modification by ACR) derived from synaptosomal proteins. These findings will provide a better understanding of the mechanisms involved with presynaptic accumulation of ACR adducts and its effect on synaptic function.

Development of a Fluorescent Affinity Tag for Phosphoprotein Detection
